When possible, take advantage of trends. Use hashes to get your message out to groups.
When you click on the 'home' link from your Twitter page, you will see the trending topics. Stay alert for topics that are relevant to what you promote.
As an example you might tweet #musicmonday (your message).
This will put your tweet into a large group that follows Music on Mondays.

Re: Follow trends
Twitter is a big popularity contest. Mathemeticians spend their time determing when you should tweet and how often based on how many friends follow you, and how many followers they have. I think it is something like [bleep] x [bleep]= [bleep].
But whatever... If Google wanted it for a billion dollars there must be some value in it. Wait a second, we all voted to give Bush a second term. hmmm... maybe there is no value.
Does anyone get results with that thing. Show me some proof, damn it!

Re: Follow trends
Its hard to gauge results from twitter, but it is undoubtedly a great way to build a fan base and actually get fans to care about what your doing on a more personal level. If you can acquire a good amount of followers, then anything you feel like saying, or any event you would like to promote, can be sent directly to them and seen by everyone.
